Radiobiological rationale for RT–drug interaction
Radiosensitizing on the same axis as AKT inhibition (impaired DSB repair, ↓survival signaling), greatest in PI3K-dependent tumors (Petroni et al., Nature Reviews Clinical Oncology, 2022; Barker et al., Nature Reviews Cancer, 2015). Because repair inhibition is not tumor-selective, a theoretical increase in acute in-field mucocutaneous/GI adverse events is plausible, potentially compounded by the class effects of colitis (idelalisib/duvelisib) and pneumonitis.

Inavolisib combined with SBRT, without concurrent use of
drugs from other classes.
