Radiobiological rationale for RT–drug interaction
Radiosensitizing in HER2-overexpressing tumors (↓HER2/AKT/ERK, delayed DSB repair, ↑apoptosis/senescence) without sensitizing HER2-negative cells (Amrell et al., Cancer Cell International, 2024; Yu et al., Oncotarget, 2016). Antigen/driver-restricted sensitization limits theoretical field side-effects, though the pan-HER class effect of diarrhea and mucocutaneous adverse events could overlap with RT-related GI/mucosal reactions.
