Radiobiological rationale for RT–drug interaction
ROS1 TKIs predominantly interact additively rather than synergistically with ionizing radiation, lacking the reinforcement in cellular injury that would drive a major theoretical increase in normal-tissue side effects (Jost et al. Neoplasia 2022). Despite this favorable radiobiological profile, the class's inherent adverse events—such as pneumonitis, gastrointestinal symptoms, and neurologic events—could theoretically compound with radiation-induced inflammation in overlapping treatment fields (Desilets et al. Cancer 2025; Jiang et al. Frontier Pharmacology 2025; Barbato et al. Clinical Cancer Research 2024).
Data on this page refer exclusively to patients treated with Taletrectinib combined with SBRT, without concurrent use of drugs from other classes. Combination therapy data and pooled group-level evidence are available on the ROS1 targeting TKI group page.
