Radiobiological rationale for RT–drug interaction
Construct-dependent, often intentionally radiation-enhancing (suicide gene/prodrug, RT-inducible promoters, immune-gene delivery) (Advani et al. Journal of Clinical Oncology 2007; Vlachaki et al. International Journal of Radiation Oncology, Biology, Physics 2001; Marco et al. Cancer Cell 2026). Adverse events likewise depends on the construct—tumor-targeted/RT-inducible expression aims to spare normal tissue, whereas immune-gene approaches carry a theoretical local/systemic inflammatory adverse event risk.
