Radiobiological rationale for RT–drug interaction
Uncertain radiosensitization (topo I/senescence rationale vs. absent free-DXd effect at low dose) (Lesperance et al., Advanced Science, 2026; Vezzoli et al., Scientific Reports, 2026). As a DXd ADC it shares the class-specific ILD/pneumonitis liability that overlaps radiation pneumonitis, and (by analogy to other DXd ADCs) a theoretical rationale for enhanced CNS normal-tissue injury with intracranial RT.

Datopotamab Deruxtecan combined with SBRT, without concurrent use of
drugs from other classes.
