Radiobiological rationale for RT–drug interaction
Radiosensitizing (blocks RT-activated EGFR→RAS/ERK, PI3K/AKT repair; S-phase depletion, senescence). Because EGFR-dependent repair and proliferation also operate in normal skin, mucosa, and lung, the mechanism is not tumor-selective and provides a rationale for increased in-field radiation dermatitis, mucositis, and radiation pneumonitis (Meng et al., International Journal of Radiation Oncology, Biology, Physics, 2024; Zheng et al., International Journal of Cancer, 2026).
